The pituitary-specific regulatory gene GHF1 contains a minimal cell type-specific promoter centered around its TATA box.

چکیده
GHF-1 is a pituitary-specific transcription factor responsible for activation of the growth hormone (GH) gene. The GHF1 gene is expressed exclusively in cells of the somatotrophic lineage, and its transcription is extinguished in somatic cell hybrids. The minimal sequences required for differential transcription of GHF1 in GH-expressing and -nonexpressing cell lines and somatic cell hybrids were localized to a 15-bp region surrounding and including its TATA box. This 15-bp fragment acts as a cell type-specific promoter element and is recognized by a transcription factor present in GH-expressing cell lines. Hence, in addition to enhancers and upstream promoter elements, the TATA element (TATA box plus surrounding sequences) can be, in certain cases, an important determinant of cell-type-specific transcription.

A specific 15-bp TATA box promoter element is required for expression of a herpes simplex virus type 1 late gene.
The herpes simplex virus type 1 (HSV-1) glycoprotein C (gC) gene is a true late or gamma 2 gene in that its expression shows a strict requirement for viral DNA replication. Elements required for regulated expression of this gene were previously shown to consist of the gC TATA box, transcription start site and a large portion of the leader sequence of the gC gene. In this paper we show that tran...
متن کاملcAMP-responsive element in TATA-less core promoter is essential for haploid-specific gene expression in mouse testis
Promoters, including neither TATA box nor initiator, have been frequently found in testicular germ cell-specific genes in mice. These investigations imply that unique forms of the polymerase II transcription initiation machinery play a role in selective activation of germ cell-specific gene expression programs during spermatogenesis. However, there is little information about testis-specific co...
متن کاملThe JC virus minimal core promoter is glial cell specific in vivo.
The glial cell specificity of the human papovavirus JC (JCV), an etiologic agent for progressive multifocal leukoencephalopathy, is thought to be due to the presence of both positive and negative regulatory elements upstream of the TATA region within the JCV promoter. Here we report that the JCV minimal core promoter, containing only the TATA box and an 8-bp poly(T) region immediately upstream,...
متن کاملRet 4, a positive acting rhodopsin regulatory element identified using a bovine retina in vitro transcription system.
Previous transgenic mouse studies demonstrated that the bovine rhodopsin sequence between -222 and +70 base pairs (bp) contains a minimal promoter, which is sufficient to direct photoreceptor cell-specific expression of a lacZ reporter gene. To more fully define the DNA regulatory elements and protein factors involved in regulating rhodopsin transcription, we have developed an in vitro transcri...
متن کاملThe cellular TATA binding protein is required for rep-dependent replication of a minimal adeno-associated virus type 2 p5 element.
The p5 promoter region of adeno-associated virus type 2 (AAV-2) is a multifunctional element involved in rep gene expression, Rep-dependent replication, and site-specific integration. We initially characterized a 350-bp p5 region by its ability to behave like a cis-acting replication element in the presence of Rep proteins and adenoviral factors. The objective of this study was to define the mi...
